"Donald Trump is not capable of managing anything," said the organization's president, Sean McGarvey.
The event, held at a Washington hotel before a crowd of union members, was the latest in the battle for the votes of blue-collar workers.
Biden has been racking up union endorsements in his bid to keep his predecessor from returning to the White House.
The president and Democratic presidential nominee has said unions will help him make Trump "a loser again," and he mocked his rival's inability to pass infrastructure legislation, saying "he's never built a damn thing."
Biden has repeatedly cast his opponent as a callous businessman who reveled in firing people on his reality TV show "The Apprentice."
"He looks down on us. I mean, think about it," Biden said.
The president has portrayed his challenger as out of touch with the concerns of working people.
The building trades unions' endorsement follows similar announcements in January and March from the United Auto Workers and the United Steelworkers.
